Abstract

The major diseases of the locomotor system, that is, osteoarthritis and osteoporosis, are so-called complex diseases. These include the typical common age-related disorders such as cancer, cardiovascular disease, and diabetes and share that they are caused by hundreds of genetic factors and are characterized by interplay of genes and environment. Epidemiological research of these complex diseases has identified anthropometric, behavioral, and serum parameters as risk factors for incidence and progression of the disease. However, genetic (DNA) polymorphisms have gained considerable interest in this respect, fueled by increasingly large genome-wide association studies (GWASs).
