Abstract
This is the first analysis describing the distribution of osimertinib in various CNS compartments in patients with CNS progression. The results could further help guiding studies that focus on optimising osimertinib penetration in this difficult-to-understand body compartment. Osimertinib accumulates into the tumour tissue, which could explain its clinically relevant effectivity against CNS metastases. However, no comparison could be made with osimertinib concentrations in patients with stable CNS disease. The large interpatient variability could indicate a subset of patients with progression due to low osimertinib CNS penetration. This data underlines the importance for future studies to focus on the individual differences in osimertinib’s CNS distribution.
