Perspectives on TLR2 agonist use in the road map to HBV cure

As we will outline in this review, Toll-Like Receptor 2 (TLR2) agonists represent potential therapeutic opportunities as either vaccine adjuvants or direct treatments for chronic HBV infections (CHB). However, to define the specific contexts in which these opportunities may be realized, it is essential first to provide a short narrative on the natural history of HBV infection, the mechanisms through which the host immune system controls viral replication, the immunological pathways engaged by TLR2 signaling, and current knowledge of the interplay between HBV infection/replication and the TLR2 signaling pathway. All this will help us delineate the potential use of TLR2 agonists in the third part of this review. Perspectives on the realistic use of therapeutic combinations will finally be given.
